(2) Test tube method ① Add 1 drop of anti-D reagent to a pre-labeled test tube. ② Add another drop of about 5% of the red blood cell suspension to be tested into the test tube. Mix well. ③ Centrifuge immediately at 1000r/min for 1 min. ④ Observe the results with the naked eye. In completely agglutinated tubes, the liquid is clear, with red blood cell clots on the bottom. The cells on the bottom of the tube are lace-like, and the clots do not disperse when the test tube is tapped lightly. In completely unagglutinated tubes, the liquid surface is clear, and the blood cells sink evenly to the bottom of the tube with neat edges. When the test tube is tapped lightly with a finger, the red blood cells immediately rise like a wisp of smoke and then become a uniform suspension. In cases of slight agglutination, the liquid must be transferred to a slide for microscopic examination before making a decision. ⑤ If agglutination occurs, it means that the D antigen Rh(D) is present on the red blood cells being tested, and if there is no agglutination, it means that the red blood cells are Rh(D) positive. |
