When treated with alcohol, the long-chain fatty acids are dissolved and the permeability of the cells is increased, making the crystal violet-iodine nitric oxide synthase easily extracted by alcohol and faded, and then stained with the color of the counterstain (safranine), thus showing a bright red color. 3.
