1. First: Gram staining reaction is a key characteristic for bacterial classification and assessment. It was pioneered by Dutch doctor Gram in 1884. Gram staining can not only observe the morphology of bacteria but also distinguish all bacteria into two categories: those with purple-blue color reaction are called Gram-positive bacteria, indicated by G; those with bright red color reaction (counter-staining tone) are called Gram-positive bacteria (G-negative bacteria), indicated by G-.

2. Second: The different reactions of bacteria to Gram staining are due to the different composition and structure of their plant cells. The plant cells of Gram-positive bacteria are relatively thick (20-80nm) and are mainly composed of peptidoglycan and teichoic acid. They have only one layer of structure, the main component of which is peptidoglycan, which accounts for 50% to 90% of the physical labor of plant cells. During the coloring process, when treated with alcohol, the diameter of the porous structure shrinks due to dehydration, the permeability decreases, and the crystal violet-iodine nitric oxide synthase is retained in the cells and is not easy to fade, thus showing a purple-blue color; the peptidoglycan content in the cells of Gram-positive and negative bacteria is low, while the content of long-chain fatty acids is high. When treated with alcohol, the long-chain fatty acids are dissolved and the permeability of the cells is increased, making the crystal violet-iodine nitric oxide synthase easily extracted by alcohol and faded, and then stained with the color of the counterstain (safranine), thus showing a bright red color.

3. Third: mainly various antibiotics. However, due to the increasingly serious drug resistance, many antibiotics, such as penicillin, have lost their former glory.
