The hormone LH increases with the passage of time, forming an LH surge, and within the next 28 hours, stimulates the release of a mature egg, i.e. ovulation.

Within 24 to 48 hours before female ovulation, the luteinizing hormone (LH) in urine will reach a peak value, and the result will be positive if the ovulation test strip is used for self-test. The specific test method is generally described on the packaging of the ovulation test strip (it must be done strictly in accordance with the instructions to minimize the error). Unlike early pregnancy test strips, morning urine cannot be used, and urine samples at the same time of day should be used as much as possible. Water intake should be reduced 2 hours before urine collection, because diluted urine samples will also hinder the detection of LH peak. If the menstrual cycle is relatively regular, the test is carried out for six consecutive days, 14 days before the menstrual period (that is, the expected ovulation time), the first three days of this time plus the last three days; if the menstrual period is irregular or abnormal, the test is generally started on the third day after the menstrual period is over. Although the test strip is easy to use, due to the production process, the operator itself and other reasons, the accuracy rate is only about 75%.

When the LH in the woman's urine on the ovulation day is less than the amount applied on the test paper, no strong positive result will be detected.
