Is fasting required for estrogen test?

You need to be on an empty stomach when doing an estrogen test and cannot eat for eight hours before the test. In addition, women should not have sexual intercourse for three days before the test. Estrogen is a common test item. Checking the secretion of estrogen is of great significance to women's health. For example, ovarian health and so on require such examination and diagnosis.

Estrogen precautions

(1) Both the labeled and standard substances are prepared with anhydrous ethanol, which is volatile. The lids should be tightly sealed and stored in a refrigerator at 4°C.

(2) After the antiserum is diluted into a mother solution, it should be stored at 4°C. Do not store it at -20°C to avoid repeated freezing and thawing, which will reduce the titer of the antiserum.

(3) Reagents such as gelatin, activated carbon, and dextran should be kept in a cool and dry place.

(4) 0.1% GPS can easily deteriorate even at 4°C in summer. It is best not to dose too much at one time. If obvious deterioration such as turbidity, odor, etc. is found, the seeds should be re-breeded.

(5) When performing B and F separation, since DCC needs to maintain a constant contact adsorption time with the sample, the number of sample tubes in a batch should not be too large, and the time should not be too long. The addition should be completed within 3 to 5 minutes; after centrifugal separation, the second batch can be added.

(6) When absorbing activated carbon solution (DCC), it is necessary to ensure that the DCC solution is fully mixed to avoid affecting the separation effect due to uneven DCC concentration.

(7) CH2Cl2 used for extraction must be redistilled before use.

Estrogen test process

1 ml of plasma sample was extracted with 0.2 ml of 0.1 mol/L NaOH and 6 ml of CH2Cl2, and the CH2Cl2 layer was washed twice with distilled water, 1 ml of distilled water each time. Pipette out two 2 ml portions of extract and place them in small test tubes, add 50 μl of 3H-estrone to each tube, and blow dry with compressed air in a 45°C water bath. Add 0.2 ml of antiserum to each tube, shake well, and place at 4°C overnight. The next day, add 0.1 ml of 5 g/L GPS, and then add 0.5 ml of 2.5 g/L DCC in a 4°C water bath. Allow to stand for 15 minutes, centrifuge for 10 minutes (3000 r/min), aspirate 0.4 ml of the supernatant and transfer to 8 ml of scintillation fluid for counting and measurement.
